The first assassination mission that Franklin receives from Lester is required to move the story forward, and it is the only one that should be completed before the final heist has been pulled off.
Kill the target using a sniper rifle.
If you want to make the most out of the assassination missions, you’ll want to make sure you have an ample amount of money saved up for each character. With Trevor you can complete the Bail Bond missions – return everyone alive for more cash – as well as finish up the Vinewood Souvenir side missions. Likewise you can purchase the Sonar Collections dock with either Franklin or Michael, completing the quests that it unlocks to earn over $900K.
Make sure it’s a Monday before you start the mission, then invest all of each character’s money into Betta Pharmaceuticals via the BAWSAQ site.
After the introduction, grab yourself some wheels and head for the objective. Once you reach the marker, move to the top floor of the garage and park along the corner looking down on the hotel entrance. Now grab a Sniper Rifle, climb onto your car, then wait for the target to exit the building on the right. Shoot him before he reaches vehicle.
Now it’s time to make your escape. Hop into your car, aim for the ramp on the other side of the garage roof, veering left and trying not to crash on the way down. Once you’re far enough away from the scene of the crime the mission will end.
Continue sleeping (saving the game) with Trevor until the return is at 80 percent, then sell Betta Pharmaceuticals.
Once you’ve sold off the Betta Pharmaceuticals stock, purchase as much as you can in Bilkinton Research. Have Trevor sleep two or three days away, then sell it at about a 50 percent return.
For the second assassination mission, Lester will task Franklin with taking down four targets within a time limit of nine minutes.
Kill the targets as quickly as possible.
Complete the mission on a Monday. Invest Franklin, Michael and Trevor’s money into Debonaire stocks on the Liberty City National Exchange (LCN).
Following the introduction, grab a car and drive to the scene of your first murder. You’ll be looking for a body builder who resembles Rocky Balboa. Once you spot him, put a round through his temple and get back in the vehicle.
The next John Doe to hit the morgue because of your bullets is up north, hanging out on a yacht beside the coast. Follow the route on your map, then locate him using the scope on your Sniper Rifle. Once you have him in your sights, drop him like a hot potato and continue on.
The third target is a bit higher up than the others, and you’re going to want to have a long range weapon ready. We used a Sniper Rifle for most of these kills, and we recommend you do the same. Head for the marker on your map. The target is riding a window cleaning lift in West Vinewood. Once you’re close enough, hop out of your car, bring up your scope, shooting him in the back to finish him off.
The final target is the toughest of them all depending on what kind of vehicle you happen to be driving. Either way, grab something fast and head toward the moving marker on your map. Once you’re close enough, ram the guy and try to run him over. If he manages to live, hop out and execute him with your pistol, mumbling something Norman Reedus like from The Boondock Saints.
Keep sleeping and checking the stock market until the return percentage is at about 80. Once all three characters are free of Debonaire, grab up as much Redwood Cigarettes stock as you can, repeating the sleep process until players get about a 300 percent return on their investment.
The third assassination Franklin is tasked with has two outcomes: A) You’ll blow the target to tiny pieces and take part in a glorious police pursuit, or B) A well-placed Sniper Rifle round will splatter his brains on the prostitute he's chatting with and you’ll escape without being seen. If you want to earn Gold Completion, you’ll need to take the second route.
Complete the mission without alerting the LSPD.
Make sure it’s Monday again, then invest all your money into Fruit Computers on BAWSAQ.
This assassination is about as easy as the first. Simply bring a silenced Sniper Rifle and you’re ready to go. Drive to the location on your map to find the working girl you need to watch, then park your car on the hill across from her location. Now it’s time to be patient and wait for your guy.
The target this round will be driving a convertible. For us it was a Peyote style vehicle. You’ll know who he is when the girl starts to get in. You don’t have much time to line up your shot, so get his head in the middle of your scope and pull the trigger. Now hop back in your car and drive out of the area to end the mission.
Play the sleeping game with Trevor until you see a 50 percent return, selling the Fruit Computers stock, then investing in Façade, finally selling it for about a 33 percent return.
So far, most of these assassination missions focused on taking out the scum of Liberty City’s elite, and this assignment is no different. Upon answering the pay phone for this mission, Lester will speak of Issac Penny, a tight-fisted billionaire planning to buy into Vapid. The real kicker here is the fact that Penny rides the bus to and from work every day. To complete this mission, you’ll need to become the bus driver and take down the iron-fist tyrant, Issac Penny.
Kill the target by hitting him with the bus.
As always, make sure it’s Monday, but do not make any investments.
Per the usual, grab yourself a car after you watch the opening cut scene, then head to the marker on your map. Once you reach the end of the line you’ll notice a bus parked in the depot. Hop into it and begin your first day as an average Joe.
Follow the bus route until a cut scene plays out between Penny and Franklin. Once the target hops off the bus, he’ll grab a nearby bike and try to outrun you. Simply run him down, making sure the bus kills him to earn Gold Completion. Exit the area to finish the mission.
Pull up the BAWSAQ site and invest all your money into Vapid stocks, waiting a few days and selling them off for about a 100 percent return.
The final of the five assassination missions is perhaps the longest and toughest. Franklin is tasked with taking down Enzo Bonelli, a man with organized crime connections who is hiding behind his armed guards at a construction site located in Pillbox Hill. In order to complete this assassination, you’ll have to fight through a dozen or so thugs and take out a helicopter. You also get to escape in style by diving off the top of the construction site and gracefully parachuting back to the ground. There’s nothing like a daring escape.

Make sure it’s Monday, then invest your money into Gold Coast stocks on the LCN.
Once you’re back in control, grab a vehicle and head to the marker on your map to spark a cut scene introducing Enzo. Once the scene ends, keep driving around to the back of the construction site. Line your vehicle up with the lone guard patrolling the dirt parking area, then run him over.
Once you’re out of the car, head for the elevator right up the path from you and take out the nearby guard with a silenced weapon. When he’s dead, hop on the elevator and ride it up to the first floor.
This floor has eight total enemies – six throughout, and two more who ride the next elevator down. Take your time through this area, using cover and making use of grenades to drive the enemies from hiding. Once they’re all dead, approach the final elevator and head up to the top floor.
This is your last stop. Here you’ll need to take out Enzo as well as any guards who get in your way. This is also where your RPG will come in handy. As you exit the elevator, shoot the guards closest to you and then take cover behind the boards ahead. Pull out your RPG and aim it at the helicopter. Make sure your aim is true and then let the rocket fly to take the target out.
To make your daring escape, head towards the parachute on the ground (unless you already have one) and then skydive off the corner of the building. We used the nearby parking garage as a landing zone, so feel free to do the same. Once your feet are back on the ground, Franklin will give Lester a call to report back that the assignment is complete.
If necessary, playing the sleeping game with Trevor until the Gold Cost stock is showing a return percentage of about 80, then sell it off. If gamers have played their cards right they should be sitting on about $2.1 billion with each character. For the record, that's about 2,100 Truffade Adder's for each character, and they go for $1 million a pop.
