How do you make a recruit an assassin on assassins creed brotherhood
[Short version - Locate a Bortia Tower, defeat the commmander without being detected by him or other guards. Climb to the top of the tower and ignite it. Find a civilian using you map, defeat the guards to recruit he/she. Find a pigeon coop or access Eztio's hideout. Here you will see your recruits and send them on contracts to raise their experience and earn you money.]

Long version (recommended if you are new to the game) Firstly, you must find a Bortia tower, this can be found throughout Roma, go to your map for assistance.
Secondly, you must kill the Bortia Commander, but the tricky part is to do this undetected by him. Some people recommend using the hidden gun from a safe distance, but this option is not always available and is sought of lame. The easiest way is to defeat guards that stop you from reaching the commander undetected. Once you kill him, the guards will either prepare to fight you or run away, so just be ready.

Thirdly, once the commander is dead, the Bortia tower is now available to be ignited. All you need to do is climb to the top and press (B). This will start a short clip of the tower exploding and Etzio leaping off the tower.

Now that the Bortia influence is gone in that area, by using your map, you can locate
a civilan that you need to protect from Bortia guards. Once you defeat the guards, speak to the civilian by pressing (y) and he/she will join you, then walk off.

Now locate a pigeon coop, or go to Eztio's hideout, there you will find a recruit added to your brotherhood. Now you can send them on contracts to raise their experience and level them up. Every level completed adds a skill point that you can use to improve the recruits abilities.

You must repeat these steps to obtain a recruit. Dont worry, it is worth it. Recruits are handy for defeating targets and earning money from contracts that you can spend on weapons, clothing, medicine etc.
